A novel method to optimize culture conditions for biomass and sporulation of the entomopathogenic fungus Beauveria Bassiana IBC1201 BJM

Biomass yields and sporulation of Beauveria bassiana was concerned on culture conditions, environmental factors and cultivation method. We optimized the best culture conditions for biomass yields of B. bassiana IBC1201 with the novel "two-stage" cultivation method as well as orthogonal matrix method. Firstly, we cultured spore suspension on the basal medium (sucrose 19.00 g, soy peptone 4.06 g, K2HPO4 1.00 g, KCl 0.50 g, MgSO4 0.50 g, FeSO4 0.10 g and 17.00 g Bactor) for the first stage culture of 4 days under room condition. Concomitant production of two proteases and alpha-amylase by a novel strain of Bacillus subtilis in a microprocessor controlled bioreactor BJM

We describe the simultaneous production of Bacillus subtilis based proteases and alpha amylase using a computer controlled laboratory scale 7.5 L batch bioreactor. The present strain is the first to be reported that concomitantly produces these two industrially important enzymes. The growth and sporulation of Bacillus subtilis was monitored and maximum production of alkaline protease and alpha amylase was found to coincide with maximum sporulation. Two types of proteases were detected in the fermentation broth; a neutral and an alkaline protease most active in a pH range of 7.0-8.0 and 8.0-10, respectively. ABSTRACT: Cassava (Manihot esculenta Crantz) is an important crop in Brazil and Pará is the major producer of roots. High temperature and humidity of tropical regions favor the development of various diseases, among them the cassava root rot. The objective of this study was to evaluate the effect of luminosity and culture medium on the mycelial growth and sporulation of Phytopythium sp. associated with different methods of inoculation on cassava roots. Control of anthracnose in green pepper involves the use of resistant varieties and/or fungicides. The selection of varieties and efficient products demands great amounts of conidia as inoculum. L’étude de l’émission de carpospores de deux espèces de gracilaires : Gracilaria gracilis et de Gracilaria bursa-pastoris a été réalisée sur des fragments de thalle femelle à cystocarpes. Ces fragments ont été prélevés à différents niveaux de la plante mère (apex, parties intermédiaires et basales) reflétant un degré plus ou moins avancé de l’âge de la plante femelle à cystocarpes. Expression of the bacterial recA gene impairs genetic recombination and sporulation in a Saccharomyces cerevisiae diploid strain Genet. Mol. Biol.

The Escherichia coli RecA protein (RecAp) has been demonstrated to induce mutagenesis in yeast cells, although there is still little information on the role of the RecAp in yeast recombination events. We evaluated spontaneous and induced general recombination in vegetative and meiotic cells of the XS2316 strain of the yeast Saccharomyces cerevisiae bearing the recA gene. We found that RecAp decreased reciprocal recombination, gene conversion and intrachromosomal recombination and promoted an increase in error-prone processes in both vegetative and meiotic cells, while its negative effect on meiotic recombination blocked ascospore formation.

Morphological changes and growth of filamentous fungi in the presence of high concentrations of PAHs BJM

In this study, we evaluated the effect of low and high molecular weight polycyclic aromatic hydrocarbons (PAHs), i.e., Phenanthrene, Pyrene and Benzo[a]pyrene, on the radial growth and morphology of the PAH-degrading fungal strains Aspergillus nomius H7 and Trichoderma asperellum H15. The presence of PAHs in solid medium produced significant detrimental effects on the radial growth of A. nomius H7 at 4,000 and 6,000 mg L−1 and changes in mycelium pigmentation, abundance and sporulation ability at 1,000–6,000 mg L−1. In contrast, the radial growth of T. asperellum H15 was not affected at any of the doses tested, although sporulation was observed only up to 4,000 mg L−1 and as with the H7 strain, some visible changes in sporulation patterns and mycelium...

Optimization of nutritional requirements for mycelial growth and sporulation of entomogenous fungus Aschersonia aleyrodis Webber BJM

The objective of the present study was to investigate the optimal nutritional requirements for mycelial growth and sporulation of entomopathogenic fungus Aschersonia aleyrodis Webber by orthogonal layout methods. Herein the order of effects of nutrient components on mycelial growth was tryptone > Ca2+ > soluble starch > folacin, corresponding to the following optimal concentrations: 1.58% Soluble Starch, 3.16% Tryptone, 0.2 mmol l-1 Ca2+ and 0.005% Folacin. The optimal concentration of each factors for sporulation was 1.16% lactose, 0.394% tryptone, 0.4 mmol l-1 Fe2+ and 0.00125% V B1, and the effects of medium components on sporulation were found to be in the order lactose > V B1 > Fe2+ > tryptone. Production and infectivity of inoculum of arbuscular mycorrhizal fungi multiplied in a substrate supplemented with Tris-HCl buffer BJM

The effect of adding Tris-HCl buffer on production and infectivity of AMF inoculum was investigated. Sporulation of Glomus etunicatum, Acaulospora longula and Gigaspora albida was improved in solution with buffer. The infectivity of G. etunicatum increased after storage, what suggests that the inoculum of this isolate is benefited by storage.

The life cycle of Haplosporidium sp., Haplosporida recently observed in the flat oyster of the bassin d'Arcachon (France), and the spore characteristics of which have already been presented (Pichot et al., 1979), has been studied by electron microscopy. The first stages of sporUïation are represented by two types of plasmodia. After the stage of sporonts, sporogenesis seems to develop according ,~ to two sequences.
